The goal of this project was to sequence invasive Bigheaded Carp genomes for downstream utilization in the development of biocontrol methods. Whole genome shotgun sequence data for Mylopharyngodon piceus assembled to Cyprinus carpio (Accession number (GCA_000951615.2). M. piceus tissue was extracted using a phenol chloroform protocol and genomic DNA was sheared to 350bp using a Covaris ME220 Sonicator. An Illumina TruSeq PCR free library preparation protocol was followed and library quality and quantity was measured using a KAPA Quantification reaction and a TapeStation 4200. The library was PE 150 sequenced using a NextSeq 500. Data was quality checked using BBDuke and assembled to the C. carpio reference genome using Geneious vs. 9.1

本项目旨在对入侵性大头鲤(Bigheaded Carp)进行基因组测序,以支撑后续生物防控方法的开发。针对青鱼(Mylopharyngodon piceus)的全基因组鸟枪测序数据,已基于登录号为GCA_000951615.2的鲤鱼(Cyprinus carpio)参考基因组完成组装。实验采用酚-氯仿法提取青鱼组织样本,使用Covaris ME220超声波破碎仪(Covaris ME220 Sonicator)将基因组DNA片段化至350bp。随后采用Illumina TruSeq无PCR文库制备流程构建测序文库,并通过KAPA定量反应试剂盒与TapeStation 4200毛细管电泳仪对文库的质量与浓度进行检测。该文库通过NextSeq 500测序平台完成PE150双端测序。测序数据使用BBDuk进行质量控制,并通过Geneious 9.1版本软件比对至鲤鱼参考基因组。
