To study genes involved in cellular response to Influenza A virus infection, human cells MRC5, WI-38 VA-13, A549 and HEK293FT were infected with influenza A/Puerto Rico/8/1934 (H1N1) virus. RNA-seq analysis of infected cell lines after 48 h was performed on an Illumina NextSeq 500 platform. The same mock-infected cells were used as controls. PolyA RNA-enriched fraction was used for constructing of cDNA libraries. Differential expressed genes were identified using R package DESeq2.

为研究参与甲型流感病毒(Influenza A virus)感染细胞应答的基因,本研究将人源细胞系MRC5、WI-38 VA-13、A549及HEK293FT以甲型流感病毒A/Puerto Rico/8/1934(H1N1)进行感染。于感染后48小时,采用Illumina NextSeq 500测序平台对感染细胞系开展RNA测序(RNA-seq)分析;以相同条件下的模拟感染细胞作为对照。实验采用聚腺苷酸RNA富集组分(PolyA RNA-enriched fraction)构建互补脱氧核糖核酸(cDNA)文库,并借助R软件包DESeq2鉴定差异表达基因。
