SrfJ is an effector of the type III secretion systems of the Gram-negative intracellular pathogen Salmonella enterica serovar Typhimurium. To study the effects of this effector on global gene expression in host cells, we have expressed SrfJ on human HeLa cells through transient transfection. The comparison with HeLa cells transfected with a plasmid not expressing SrfJ, revealed a number of genes that are differentially expressed when SrfJ is present. Overall design: Two conditions. Each condition replicated three times. Condition 1: control of HeLa cells transiently transfected with plasmid pcDNA3 without SrfJ. Condition 2: HeLa cells transiently transfected with plasmid pcDNA3 with SrfJ.

SrfJ是革兰氏阴性胞内病原体鼠伤寒沙门氏菌(Salmonella enterica serovar Typhimurium)III型分泌系统(type III secretion system)的效应蛋白(effector)。为探究该效应蛋白对宿主细胞全局基因表达的影响,我们通过瞬时转染技术在人源HeLa细胞中表达SrfJ。将该实验组与转染不含SrfJ的pcDNA3质粒的HeLa细胞进行对照分析,可鉴定得到一批在SrfJ存在时呈现差异表达的基因。 实验设计概述:设置两组实验条件,每组均进行三次生物学重复。条件1:转染不含SrfJ的pcDNA3质粒的HeLa细胞(对照组);条件2:转染携带SrfJ的pcDNA3质粒的HeLa细胞。
