We established a novel epigenomic profiling method ChIL-seq which does not employ immunoprecipitation, but instead exploits immunostaining. We performed ChIL-seq on C2C12, NIH3T3, K562, MCF-7 and MDA-MB-231 cells with various cell numbers (1, 100, 1k, 10k and 100k cells) against various histone modifications and transcription factors, as well as H3K4me3 ChIP-seq for C2C12, MCF-7 and MDA-MB-231.

本研究建立了一种新型表观基因组谱分析方法——染色质整合标记测序(ChIL-seq),该方法无需依赖免疫沉淀(immunoprecipitation)技术,而是通过免疫染色(immunostaining)完成实验流程。研究团队针对多种组蛋白修饰与转录因子靶标,在C2C12、NIH3T3、K562、MCF-7及MDA-MB-231细胞系中开展了ChIL-seq实验,所用细胞梯度包含1个、100个、1000(1k)、10000(10k)及100000(100k)个细胞;同时还针对C2C12、MCF-7与MDA-MB-231三种细胞完成了H3K4me3染色质免疫沉淀测序(ChIP-seq)实验。
