Purpose: The goal of this study was to compare the mRNA transcriptomes of transgenic pigs and wild-type pigs for five organs (liver,longissimus dorsi,psoas major,subcutaneous fat and retroperitoneal fat). Methods: Total RNA was extracted from longissimus dorsi, psoas major, subcutaneous fat, retroperitoneal fat and liver using TRIZOL reagent (Life Technologies, Beijing, China), and each tissue has three biological replicates. For RNA sequencing, approximately 2 ug total RNA of each sample was used to construct transcriptome libraries following the manufacture’s standard procedures. Libraries were sequenced via Illumina HiSeqTM 4000, and 100 bp paired-end reads were generated. Results: We detected 18,051 protein coding mRNA transcripts across 30 samples . Conclusions: Human FST induced transcriptome changes in liver, Overall design: Five organs (liver, longissimus dorsi, psoas major, subcutaneous fat and retroperitoneal fat) mRNA profiles of transgenic pigs and wild type pigs were generated by deep sequencing, in triplicate using Illumina HiSeqTM 4000.
研究目的:本研究旨在比较转基因猪与野生型猪五个器官(肝脏、背最长肌、腰大肌、皮下脂肪及腹膜后脂肪)的mRNA转录组。方法:采用TRIZOL试剂(Life Technologies,中国北京)从背最长肌、腰大肌、皮下脂肪、腹膜后脂肪及肝脏中提取总RNA,每份组织设置3个生物学重复。针对RNA测序,取每份样本约2μg总RNA,按照试剂盒标准流程构建转录组文库。文库通过Illumina HiSeqTM 4000进行测序,生成100 bp双端读段。结果:本研究在30个样本中共检测到18051个编码蛋白质的mRNA转录本。结论:人源卵泡抑素(FST)诱导了肝脏内的转录组变化。整体实验设计:采用Illumina HiSeqTM 4000对转基因猪与野生型猪的五个器官(肝脏、背最长肌、腰大肌、皮下脂肪及腹膜后脂肪)进行深度测序,每份样本设置3次生物学重复,获取其mRNA表达谱。
