本研究采用高通量技术,旨在鉴定信使RNA(messenger RNA, mRNA)的表达水平,以解析本土猪与杂交猪的外周血单个核细胞(peripheral blood mononuclear cells, PBMCs)及巨噬细胞(Macrophages)中猪瘟(Classical Swine Fever, CSF)的比较发病机制。整体实验设计:分别于接种猪瘟疫苗病毒后第7天、第21天采集杂交猪与本土猪的外周血单个核细胞;同时在强毒猪瘟病毒感染杂交猪与本土猪的单核细胞衍生巨噬细胞(monocyte-derived macrophages, MDMs)60小时后采集样本,以此鉴定全基因组的基因表达情况。
