The GFP or ChREBP were overexpressed in the liver of C57Bl6/J mice through adenoviral gene delivery. Mice were studied in the fed state 3 weeks later. We used microarray analysis to detail the global program of gene expression underlying ChREBP overexpression and identified distinct classes of up and down regulated genes. Overall design: RNA extraction and hybridization on Affymetrix microarrays.

通过腺病毒基因递送技术，在C57Bl6/J小鼠的肝脏中过表达绿色荧光蛋白（Green Fluorescent Protein, GFP）或碳水化合物反应元件结合蛋白（Carbohydrate Response Element Binding Protein, ChREBP）。于转染3周后，在进食状态下对上述小鼠开展实验研究。本研究采用基因芯片（microarray）分析技术，详细解析了ChREBP过表达所介导的全局基因表达程序，并鉴定得到多个不同类别的上调基因与下调基因。总体实验设计：提取各组总RNA，随后在Affymetrix基因芯片上完成杂交实验。
