Revealing the virome of Ixodes ricinus ticks from northern Europe

RNA/transcriptome sequencing was performed on six libraries consisting of 33 Ixodes ricinus nymphs and adults sampled from six localities in Norway. For all libraries, ribosomal RNA (rRNA) was removed using the Ribo-Zero Gold (epidemiology) Kit (Illumina) following the manufacturer’s instructions. Libraries were constructed for all rRNA-depleted RNA-samples using the KAPA Stranded RNA-Seq Kit following the manufacturer’s instructions. Sequencing libraries were pooled in equimolar amounts. All libraries were sequenced on a single lane (paired-end, 125bp read-length) on an Illumina HiSeq 2500 platform

本研究对采自挪威6个采样位点的33只蓖麻硬蜱（Ixodes ricinus）若虫与成虫样本构建的6个文库开展RNA/转录组测序（RNA/transcriptome sequencing）。所有文库均按照制造商说明书，使用Illumina公司的Ribo-Zero Gold（流行病学）试剂盒去除核糖体RNA（rRNA）。针对经核糖体RNA去除的全部RNA样本，依据制造商操作指南，采用KAPA Stranded RNA-Seq Kit完成文库构建。将所有测序文库以等摩尔浓度混合。全部混合文库均在Illumina HiSeq 2500测序平台的单个测序泳道上完成双端测序，读长为125bp。
