In this study, we investigated LINE1 (L1), a major family of transposon elements (TEs) in the human genome, in naive hES cells for its roles in regulating 8-cell (8C) stage genes expression. Overall design: PXGL media maintained naive H9 hES cells were transfected with L1 targeting sense oligos (SO-L1) and anti-sense oligos (ASO-L1), respectively. Transfected cells were maintained in the PXGL media. 48 hours later, cells were directly processed for RNA extraction and RNA-seq library prep.

本研究以人类基因组中主要转座因子(transposon elements, TEs)家族之一的长散在核元件1(LINE1, L1)为研究对象,在幼稚态人类胚胎干细胞(naive hES cells)中探究其对8细胞(8C)阶段基因表达的调控作用。实验整体设计:将维持于PXGL培养基中的H9幼稚态人类胚胎干细胞分别转染靶向LINE1的正义寡核苷酸(sense oligos, SO-L1)与反义寡核苷酸(anti-sense oligos, ASO-L1)。转染后的细胞继续培养于PXGL培养基中,转染48小时后直接收集细胞进行RNA提取与RNA测序(RNA-seq)文库制备。
