In this study the effect of anti-miR-33 treatment on plasma and liver lipid profiles was examined in non-human primates. A significant increase in plasma HDL-C, and a significant decrease in plasma triglyceride levels were observed. African green monkeys on a Normal chow diet were treated with anti-miR-33 or control anti-miR for 4 weeks and were then switched to a High Carb/Med. Cholesterol diet and treated with anti-miR-33 or control anti-miR for 8 more weeks (n=6/group). Gene expression profiling was performed on liver biopsies obtained at -5 weeks (baseline), 4 weeks and 12 weeks.

本研究以非人灵长类动物为实验对象,考察了抗miR-33 (anti-miR-33) 治疗对血浆与肝脏脂质谱的影响。研究观察到血浆高密度脂蛋白胆固醇(HDL-C)水平显著升高,血浆甘油三酯水平显著降低。实验中,非洲绿猴先以普通饲料日粮饲养,同时分别施加抗miR-33或对照抗miR处理,持续4周;随后将其转换为高碳水/中等胆固醇日粮,继续施加对应处理8周,每组动物数量为6只(n=6/group)。分别在第-5周(基线期)、第4周及第12周采集肝脏活检样本,开展基因表达谱分析。
