Aging is a complex biological process accompanied with time-dependent functional decline that affects most living organisms. Several interventions, including rapamycin, senolytics, NAD precursors, sirtuin-activating compounds, metformin, exercise and calorie restriction, are found to potentially increase healthspan and/or lifespan. However, pivotal methods to intervene aging are still in deficiency, because global view of natural aging is far from completeness by now. Omics analysis has the advantage to deliver an overall profile for biological processes. Liver is the biggest metabolic organ in mammals, but its global alterations during natural aging is unclear. Hereby, we performed transcriptome analysis with the liver of differentially aged mice under physiological conditions to obtain a fundamental dataset for future aging research.

衰老是一种复杂的生物学过程,伴随随时间推移的功能衰退,可影响绝大多数生命体。目前已发现雷帕霉素(rapamycin)、衰老细胞清除剂(senolytics)、烟酰胺腺嘌呤二核苷酸(NAD)前体、沉默信息调节因子激活剂(sirtuin-activating compounds)、二甲双胍、运动及热量限制等多种干预手段,可潜在延长健康寿命与/或寿命。然而,由于当前对自然衰老的全局认知仍远未完备,衰老干预的关键方法仍较为匮乏。组学分析(omics analysis)能够为生物学过程提供全面的整体表征。肝脏是哺乳动物体内最大的代谢器官,但其在自然衰老过程中的全局分子改变仍未明确。据此,本研究针对生理条件下不同年龄小鼠的肝脏组织开展转录组分析(transcriptome analysis),以期为后续衰老相关研究提供基础数据集。
