The activation of endothelium by tumor cells is one of the main steps by tumor metastasis. The role of the blood components (platelets and leukocytes) in this process remain unclear. We used microarrays to detail the repsonse of endothelial cells to tumor cells in the presence or absence of blood components. Overall design: HMVECs were cultured to 90% confluency. Tumor cells (LS180 or HT-29) in the presence or in the absence of blood components were co-cultured for 8 h with HMVECs. After the purfication of HMVECS by MACS sorting RNA was isolated.
肿瘤细胞激活内皮细胞是肿瘤转移的关键步骤之一。血液组分(血小板与白细胞)在该过程中的作用尚未明确。我们借助微阵列(microarrays)技术,详细解析了在存在或缺失血液组分的条件下,内皮细胞对肿瘤细胞的应答反应。实验整体设计:将人微血管内皮细胞(HMVECs)培养至90%汇合度。将肿瘤细胞(LS180或HT-29)分别在有、无血液组分存在的情况下,与人微血管内皮细胞共培养8小时。随后通过MACS分选(MACS sorting)纯化人微血管内皮细胞,之后提取总RNA。
