IntroductionSpodoptera frugiperda is a serious world-wide agricultural pest. Gut microorganisms play crucial roles in growth, development, immunity and behavior of host insects.
MethodsHere, we reported the composition of gut microbiota in a laboratory-reared strain of S. frugiperda using 16S rDNA sequencing and the effects of gut microbiota on the reproduction.
ResultsProteobacteria and Firmicutes were the predominant bacteria and the taxonomic composition varied during the life cycle. Alpha diversity indices indicated that the eggs had higher bacterial diversity than larvae, pupae and adults. Furthermore, eggs harbored a higher abundance of Ralstonia, Sediminibacterium and microbes of unclassified taxonomy. The dynamics changes in bacterial communities resulted in differences in the metabolic functions of the gut microbiota during development. Interestingly, the laid eggs in antibiotic treatment groups did not hatch much due to the gut dysbacteriosis, the results showed gut microbiota had a significant impact on the male reproduction.
DiscussionOur findings provide new perspectives to understand the intricate associations between microbiota and host, and have value for the development of S. frugiperda management strategies focusing on the pest gut microbiota.
引言:草地贪夜蛾(Spodoptera frugiperda)是全球性重大农业害虫。肠道微生物在宿主昆虫的生长、发育、免疫及行为调控中发挥关键作用。
方法:本研究采用16S rDNA测序技术,分析了实验室饲养的草地贪夜蛾(S. frugiperda)品系的肠道菌群组成,并探究了肠道菌群对宿主繁殖的影响。
结果:变形菌门(Proteobacteria)与厚壁菌门(Firmicutes)为该肠道菌群的优势类群,其分类组成在昆虫生活史中呈现动态变化。α多样性(Alpha diversity)指数显示,卵期的细菌多样性显著高于幼虫期、蛹期与成虫期。此外,卵期含有更高丰度的罗尔斯通氏菌属(Ralstonia)、沉积物杆菌属(Sediminibacterium)以及未分类的微生物类群。细菌群落的动态变化导致肠道菌群在宿主发育过程中的代谢功能产生差异。值得注意的是,抗生素处理组的卵因肠道菌群失调(gut dysbacteriosis)孵化率显著降低,研究结果表明肠道菌群对雄性繁殖具有显著影响。
讨论:本研究结果为解析菌群与宿主间的复杂互作关系提供了新视角,同时也为以该害虫肠道菌群为靶点的草地贪夜蛾防控策略开发提供了重要参考价值。
