Analyses of the consequences of up- and down-regulating SCL/TAL1 in human enriched hematopoietic stem progenitor cells (HSPCs). We used microarrays to detail the global gene expression program of human cord blood HSPCs (CD34+CD38-lowCD34RA-CD90+) in which SCL/TAL1 expression is either up- or down-regulated. Overall design: We have used lentiviral vector transduction of human cord blood HSPCs to overexpress (O/E) or knock down (KD) SCL/TAL1. Cells were lysed 24 hours after the end of transduction.
本数据集针对在人源富集的造血干细胞祖细胞(hematopoietic stem progenitor cells, HSPCs)中上调或下调SCL/TAL1所产生的调控效应展开分析。我们采用基因芯片技术,对SCL/TAL1表达分别被上调或下调的人脐带血HSPCs(CD34+CD38-lowCD34RA-CD90+)的全局基因表达谱进行精细解析。
实验整体设计:我们通过慢病毒载体转导人脐带血HSPCs,以实现SCL/TAL1的过表达(O/E)或敲低(KD)。转导结束24小时后,对细胞进行裂解处理。
