This study aims to identify suppression mutations in LTA deficient Staphylococcus aureus. LTA deficient Staphylococcus aureus was made by deletion of gene ltaA in strain NCTC8325. The ltaA deletion strain shows strong growth defect on Luria-Bertani (LB) agar plate low pH (pH=6.0), and colonies with restored growth show up sporadically. Whole genome sequencing of 3 colonies with restored growth and the parent ltaA deletion strain identifies suppressor mutations in each of the 3 colonies.
本研究旨在鉴定脂磷壁酸(Lipoteichoic acid, LTA)缺陷型金黄色葡萄球菌中的抑制突变。研究人员通过在菌株NCTC8325中敲除ltaA基因,构建得到LTA缺陷型金黄色葡萄球菌。该ltaA基因敲除菌株在pH为6.0的Luria-Bertani(LB)琼脂平板上表现出显著的生长缺陷,且恢复生长的菌落会零星出现。对3株恢复生长的菌落及其亲本ltaA敲除菌株开展全基因组测序后,在这3株菌落中均检测到了抑制突变。
