CRISPRi-sequencing performed on S. pneumoniae D39V in conditions where spv_0476 is either depleted or complemented, in order to identify which operons become more or less essential when silenced using CRIPRi. Illumina sequencing of the sgRNAs targeting all genetic elements after selection reflects directly the abundance of each sgRNA and thus the essentiality of an operon.

本数据集采用CRISPR干扰测序(CRISPRi-sequencing)技术,以肺炎链球菌(S. pneumoniae)D39V菌株为研究对象,在spv_0476基因分别处于耗竭或互补表达的条件下开展实验,旨在鉴定经CRISPRi技术沉默后,哪些操纵子的必需性出现增强或减弱的变化。对筛选后靶向所有遗传元件的单引导RNA(sgRNA)进行Illumina测序(Illumina sequencing),所得结果可直接反映每种sgRNA的丰度,进而体现对应操纵子的必需性。
