DNMT1 plays a major role in embryonic development as a maintenance methyltransferase. Although recent studies have shown that DNMT1 has de novo methylation activity, the detailed role of its function during embryonic development remains unclear. In this study, to further understand the role of DNMT1 de novo methylation, we performed RNA-seq and DIP-seq on DNMTs mutatnt mESCs. Overall design: RNA-seq was performed on TKO, TKO+FL and TKO+602 mESCs. DIP-seq was performed on TKO_FL and DKO mESCs.
DNA甲基转移酶1 (DNMT1) 作为维持性甲基转移酶,在胚胎发育过程中发挥核心作用。尽管近期研究表明DNMT1具备从头甲基化(de novo methylation)活性,但其在胚胎发育中的具体功能仍未明确。本研究为进一步阐明DNMT1从头甲基化的功能角色,对DNA甲基转移酶家族(DNMTs)突变型小鼠胚胎干细胞(mESCs)开展了RNA测序(RNA-seq)与DNA免疫沉淀测序(DIP-seq)实验。整体实验设计如下:针对TKO、TKO+FL及TKO+602型小鼠胚胎干细胞开展RNA-seq实验;针对TKO_FL与DKO型小鼠胚胎干细胞开展DIP-seq实验。
