Presetting of adaptive hematopoiesis by Fli-1 entails reciprocal niche sensing of Notch/VEGF-A pathways to supersede Spi1-driven dormancy. RNA-seq analysis for WT and Fli-1 KO HSPCs was performed to uncover changes in transcriptional output following Fli-1 deletion. RNA sequencing of WT and Fli-1 KO HSPCs after 6d in co-culture with a vascular niche post Fli-1 deletion induction.

Fli-1介导的适应性造血预先设定,需通过对龛微环境中Notch与VEGF-A(血管内皮生长因子A,Vascular Endothelial Growth Factor A)通路的双向信号感知,以取代Spi1驱动的细胞休眠状态。本研究针对野生型(Wild Type, WT)与Fli-1基因敲除(Knock Out, KO)的造血干细胞与祖细胞(Hematopoietic Stem and Progenitor Cells, HSPCs)开展RNA测序(RNA-sequencing, RNA-seq)分析,以揭示Fli-1缺失后转录输出的变化情况。在诱导Fli-1缺失后,将上述细胞与血管龛共培养6天,随后对其进行RNA测序。
