HIV-1 infection of monocyte-derived macrophages does not elicit a detectable type I IFN response in vitro, however, previously published data has shown that blocking STAT1 and STAT3 inhibits HIV-1 replication. Here we test to see if low levels of IFN inducible genes are detectable in human monocyte-derived macrophages that have been infected with HIV-1 in vitro. Overall design: Monocytes from 5 healthy human donors were differentiated into macrophages in vitro and infected with HIV-1-AD for 7 days. Total RNA was isolated and gene expression was assessed.

体外(in vitro)环境下,单核细胞衍生巨噬细胞(monocyte-derived macrophages, MDM)感染人类免疫缺陷病毒1型(HIV-1)后,无法触发可被检测到的I型干扰素(type I IFN)应答。但既往发表的研究数据表明,阻断信号转导与转录激活因子1(signal transducer and activator of transcription 1, STAT1)与信号转导与转录激活因子3(signal transducer and activator of transcription 3, STAT3)可抑制HIV-1的复制。本研究旨在探究体外感染HIV-1的人类单核细胞衍生巨噬细胞中,是否可检测到低水平表达的干扰素诱导基因(IFN-inducible genes)。实验整体设计:从5名健康人类供体体内分离单核细胞,经体外诱导分化为巨噬细胞后,以HIV-1-AD毒株感染细胞并培养7天;随后提取总RNA,对基因表达水平进行检测分析。
