本研究开发了一种基于补骨脂素探针(psoralen probe, PP)的RNA标记与RNA-蛋白质复合物(RNA-protein complex, RP-complex)富集方法,通过该PP富集策略可从海拉细胞中分离得到编码RNA与非编码RNA。通过RNA测序(RNA-sequencing)分析,探究PP分离所得RNA的类型与相对分布特征。同时对经放线菌素D(ActD)处理、处于转录抑制状态下的RNA进行动态监测。整体实验设计:分别通过PP富集法与TRIzol法提取海拉细胞中的RNA,分析其RNA类型与相对分布;并检测经ActD处理的海拉细胞的RNA读段计数。
