Effect of SUB1 knockdown on aggressive prostate cancer

SUB1 is known to be playing critical role in vital cellular processes such as DNA replication, repair and heterochromatization. Expression profiling studies in several cancers show its overexpression. However, its regulation and function in cancer has been less explored. Our study has indicated high expression of SUB1 in aggressive prostate cancer, we also found that the knockdown of SUB1 led to alterations in cell proliferation, invasion and migration. In addition, we found that mir-101, a tumor suppressor microRNA targets and regulates the expression of SUB1 Two-color experiment.
已知SUB1在DNA复制、修复及异染色质化等关键细胞进程中发挥核心作用。针对数种癌症的表达谱研究均显示,SUB1存在过表达现象。然而,目前针对SUB1在癌症中的调控机制与生物学功能的研究仍较为匮乏。本研究证实,SUB1在侵袭性前列腺癌中呈高表达状态;同时发现,敲低SUB1可导致细胞增殖、侵袭及迁移能力发生显著改变。此外,本研究通过双色实验发现,作为肿瘤抑制性微小RNA(microRNA)的mir-101可靶向调控SUB1的表达。
