Cyclic GMP-AMP synthase (cGAS) is a well-known cytosolic DNA sensor that activates innate immune responses by activating STING signal, which potentially mediates protective immune defense against infection. However, the mechanisms of nuclear translocation of cGAS, and the potential functions of nuclear cGAS signaling, which seems not to be involved in cGAS-STING pathway, have remained undetermined.
