To validate the results in primary endothelial cells including HUVECs and HMVECs, the effect of HEY1 SUMOylation on angiogenic functions was examined in MLECs isolated form Hey1 iEC-KO HEY1-WT/3KR iEC-KI mice. Overall design: Samples were MLECs isolated form Hey1 iEC-KO HEY1-WT/3KR iEC-KI mice. Six samples were divided into two groups: HEY1-WT iEC-KI group and HEY1-3KR iEC-KI group. There are 3 replicates in each group.
为在包括人脐静脉内皮细胞(HUVECs)与人微血管内皮细胞(HMVECs)在内的原代内皮细胞(primary endothelial cells)中验证实验结果,本研究在分离自Hey1诱导型内皮细胞特异性敲除(iEC-KO)且携带HEY1野生型(HEY1-WT)/3KR敲入(iEC-KI)小鼠的小鼠肺微血管内皮细胞(MLECs)中,检测了HEY1的SUMO化修饰(SUMOylation)对血管生成功能的影响。整体实验设计如下:实验样本为分离自Hey1 iEC-KO HEY1-WT/3KR iEC-KI小鼠的MLECs;共6份样本分为两组,即HEY1-WT iEC-KI组与HEY1-3KR iEC-KI组,每组设置3个生物学重复。
