Benign prostatic hyperplasia (BPH) is a common urological disease that adversely affects quality of life among elderly males, but its etiology remains poorly understood. We show that the expression of the androgen receptor (AR) is decreased in the luminal epithelial cells of human BPH specimens and is inversely correlated with the degree of regional prostatic inflammation. To identify the extracellular signaling that promotes epithelial proliferation, we performed RNA sequencing of FACS-isolated prostate luminal cells from tamoxifen-treated control and K8-AR mice (with knockout of androgen receptor).
良性前列腺增生(Benign prostatic hyperplasia, BPH)是一类常见的泌尿外科疾病,会对老年男性的生活质量造成不良影响,但其病因学仍不甚明确。本研究发现,人良性前列腺增生标本的腔上皮细胞中,雄激素受体(androgen receptor, AR)的表达水平下调,且该表达水平与局部前列腺炎症程度呈负相关。为明确可促进上皮细胞增殖的细胞外信号转导机制,我们对经他莫昔芬处理的对照组小鼠及雄激素受体基因敲除的K8-AR小鼠的荧光激活细胞分选(Fluorescence Activated Cell Sorting, FACS)分离得到的前列腺腔上皮细胞开展了RNA测序。
