The innate immune response to 1st generation adenoviral vectors was ascertained through high level infection of primary cells from a inbred mouse strain, replete with an intact and functioning TLR system. Sixteen hrs post-infection, RNA was harvested, transcribe to cDNA and bitotin-labeled, and hybridized to Mu11k A and B arrays.本研究通过对携带完整且功能健全的Toll样受体（Toll-like receptor, TLR）系统的近交系小鼠原代细胞进行高效感染，明确了针对第一代腺病毒载体的固有免疫应答。 关键词：免疫应答 实验设计：将静息状态的小鼠C57/Bl6成纤维细胞分别以感染复数（multiplicity of infection, MOI）=300的腺病毒（adenovirus, Ad）（E1-E3-LacZ）或空白对照进行感染。感染后16小时，收集总RNA并反转录为cDNA，经生物素标记后与Mu11k A和B基因芯片进行杂交。
