Cerebral ischemia stroke (CIS) is a multifactorial and diverse neurological disorder characterized by a high mortality rate and longterm cognitive impairment, partly due to apoptosis. However, the development of neuroprotective drugs to combat apoptosis has faced significant challenges. Puerarin (Pue), an anti-antioxidant compound extracted from the root of the Pueraria lobata (Willd), shows protective effects against against cerebral ischemia/reperfusion (CIR)-induced injury. In this study, to identify DNA methylation-mediated downstream target genes regulated by Pue, we performed Whole genome bisulfite sequencing analysis on hippocampal samples from MCAO and MCAO+Pue (60 mg/kg) groups.

缺血性脑卒中(Cerebral ischemia stroke, CIS)是一类多因素驱动、表型多样的神经系统疾病,以高死亡率与长期认知功能障碍为典型特征,其发病机制部分与细胞凋亡相关。然而,针对细胞凋亡开发神经保护药物的研究仍面临诸多重大挑战。葛根素(Puerarin, Pue)是从野葛(Pueraria lobata (Willd.))根中提取的抗氧化化合物,对脑缺血再灌注(Cerebral ischemia/reperfusion, CIR)诱导的脑损伤具有明确保护作用。本研究旨在鉴定葛根素调控的、受DNA甲基化介导的下游靶基因,为此我们对大脑中动脉闭塞(Middle Cerebral Artery Occlusion, MCAO)组与大脑中动脉闭塞+葛根素(60 mg/kg)组的海马组织样本开展了全基因组亚硫酸氢盐测序分析。
