In order to compare the small RNA (sRNA) population between the control and Potato spindle tuber viroid (PSTVd) upon infecting the plants, the tomato plants (Lycopersicum esculentum cv. Rutgers) were mock inoculated. At 21 dpi,tTotal RNA was extracted and subjected for deep-sequencing using Illumina MiSeq platform. The primers were trimmed and the 21- to 24-nt long small RNA species were filtered after quality check of the raw data. Overall design: Examination for the small RNA population in tomato plants.
为比较空白对照与接种马铃薯纺锤块茎类病毒(Potato spindle tuber viroid, PSTVd)的番茄植株间小RNA(small RNA, sRNA)种群的差异,本研究以栽培品种Rutgers的番茄(Lycopersicum esculentum cv. Rutgers)为材料,分别进行模拟接种与PSTVd接种处理。于接种后21天(21 dpi)提取总RNA,采用Illumina MiSeq平台开展深度测序。对原始测序数据进行质量质控后,裁剪去除引物序列,并筛选得到长度为21至24 nt的小RNA片段。本实验整体设计旨在解析番茄植株内的小RNA种群特征。
