Microarray analysis was performed using germ cells and gonadal somatic cells isolated by Fluorescence Activated Cell Sorter (FACS) from XX and XY embryos at st.33 and st.35. Overall design: Medaka germ cells (olvas-EGFP) and somatic cells (sox9b-DsRed) were isolated by FACS. The microarray experiments were done using three independent biological replicates.

本研究以第33期(st.33)和第35期(st.35)的XX、XY型青鳉胚胎为材料,通过荧光激活细胞分选术(Fluorescence Activated Cell Sorter, FACS)分离其生殖细胞与性腺体细胞,进而开展微阵列分析。实验整体设计如下:以olvas-EGFP标记的青鳉生殖细胞与sox9b-DsRed标记的体细胞为分选对象,经FACS完成分离;微阵列实验设置3次独立生物学重复。
