Background: Cancer testis antigens (CTAs) are an extensive gene family with a unique expression pattern restricted to testis tissues, but aberrantly high expressed in a variety of cancer tissues. In view of this, CTAs are typically considered as efficient biomarkers and targets of immunotherapy. However, limited information is available regarding CTAs as immunotherapeutic targets for gastric cancer (GC) patients. Herein, we evaluated the expression levels of CTAs in both Eastern and Western GC patients and elucidate the expression pattern and functions of a novel CTAs. Overall design: Comparative gene expression profiling analysis of RNA-seq data for MKN28/MKN45 cells with knockdown and overexpressed ZNF165.
背景:癌-睾丸抗原(Cancer testis antigens,CTAs)是一类广泛分布的基因家族,具备独特的表达模式——仅局限于睾丸组织,但在多种癌组织中异常高表达。基于该特性,CTAs通常被视为高效的生物标志物与免疫治疗靶点。然而,目前关于CTAs作为胃癌(gastric cancer,GC)患者免疫治疗靶点的相关信息仍十分有限。本研究评估了东西方胃癌患者体内CTAs的表达水平,并阐明了一种新型CTAs的表达模式与功能。
总体实验设计:对ZNF165基因敲低及过表达的MKN28、MKN45细胞进行RNA测序(RNA-seq)数据的比较基因表达谱分析。
