The mechanistic basis for humoral immune control of Mtb remains poorly understood. We use single-cell RNA-sequencing to explore the impact of antibody treatment on cellular states in vivo. Overall design: Using single-cell RNA-sequencing, we profile CD45+ and infected Mtb+ cells in B6 mice 5 days post-infection following treatment with different antibodies.
目前针对结核分枝杆菌(Mycobacterium tuberculosis, Mtb)的体液免疫调控机制仍尚未完全阐明。本研究借助单细胞RNA测序(single-cell RNA-sequencing)技术,探究抗体治疗对体内细胞状态的影响。整体实验设计如下:对B6小鼠实施结核分枝杆菌感染,在经不同抗体处理后的感染后第5天,对其体内CD45阳性细胞以及结核分枝杆菌感染阳性细胞进行单细胞RNA测序分析。
