本研究在常氧(normoxia)与低氧(hypoxia)条件下培养SK-N-BE(2)细胞48小时,针对缺氧诱导因子-1α(HIF-1alpha)开展染色质免疫共沉淀测序(ChIP-seq)实验。
我们在TET1基因中鉴定出两个全新的HIF-1alpha结合位点,这些位点可在低氧条件下调控TET1的表达。
整体实验设计:将SK-N-BE(2)细胞分为常氧、低氧两组,分别培养48小时后,对固定后的染色质进行HIF-1alpha染色质免疫共沉淀实验。
