We use next-generation sequencing (NGS) to profile miRNA in small arteries of mice infused with angiotensin (Ang) II for 7 and 14 days. The goal is to identify differentially expressed miRNAs that are associated with Ang II-induced vascular injury and hypertension. Overall design: Ten to 12-week-old male C57BL/6J WT (Harlan laboratories, Indianapolis, IN) mice were infused SC with Ang II (1000 ng/kg/min) with ALZET osmotic mini pumps (model 1002, Durect Corporation) for 7 or 14 days. Control mice underwent sham surgery. At the end of the treatments, total RNA was extracted from mesenteric arteries for miRNA profiling using NGS by Illumina HiSeq 2500.
我们采用下一代测序技术(next-generation sequencing, NGS),对经血管紧张素II(angiotensin II, Ang II)持续输注7天及14天的小鼠小动脉中的微小RNA(miRNA)进行谱分析。本研究旨在筛选与血管紧张素II诱导的血管损伤及高血压相关的差异表达微小RNA。
