Here we constructed a high-quality reference genome for <i>M. rosenbergii</i> by using Nanopore, Illumina, and high-throughput chromosome conformation capture (Hi-C) technologies. The final genome size is 3.18 Gb, with the scaffold N50 and contig N50 of 62.73 Mb and 8.92 Mb, respectively. Approximately 98.6% of assembled sequence were anchored into 59 pseudo-chromosomes. Benchmarking Universal Single-Copy Orthologs (BUSCO) benchmark of genome assembly reached 94.5%. Repetitive sequences comprise 43.77% of the assembled genome, and 17,436 protein-coding genes were annotated. The high-quality genome of<i> M. rosenbergii</i> will empower molecular breeding efforts and provide invaluable resources for comparative genomic analysis of decapod species.
本研究采用纳米孔(Nanopore)测序、Illumina测序及高通量染色体构象捕获(Hi-C)技术,构建了罗氏沼虾(M. rosenbergii)的高质量参考基因组。最终组装得到的基因组大小为3.18 Gb,支架N50(scaffold N50)和重叠群N50(contig N50)分别为62.73 Mb和8.92 Mb。组装序列中约98.6%被锚定至59条伪染色体(pseudo-chromosomes)中。采用通用单拷贝同源物基准测试(BUSCO)对本次基因组组装进行评估,其完整度达94.5%。重复序列占组装基因组的43.77%,共注释得到17436个蛋白质编码基因。该罗氏沼虾高质量参考基因组将为其分子育种研究提供有力支撑,并为十足目(decapod)物种的比较基因组学分析提供宝贵的研究资源。
