RNA was isolated from yeast Magnusiomycet capitatus. Structurally unconstrained adenosines were modified by diethyl pyrocarbonate (DEPC). Modified sites were identified by comparison of DEPC-treated and control samples using direct RNA sequencing on the Oxford nanopore MinION sequencer, following a protocol by Bizuayehu et al. 2022.

本研究从头状马格努西酵母(Magnusiomycet capitatus)中提取核糖核酸(RNA)。采用焦碳酸二乙酯(diethyl pyrocarbonate, DEPC)对无结构约束的腺苷进行化学修饰。参照Bizuayehu等人2022年发布的实验方案,使用牛津纳米孔MinION测序仪(Oxford nanopore MinION sequencer)开展直接RNA测序,通过对比DEPC处理组与对照组样本,实现修饰位点的鉴定。
