Artificial miRNA mediated knock-down ago18 transgenic lines [ago18(1), ago18(2)] showed developmentally compromised phenotype in both vegetative and reproductive tissues compared to wild-type control. We have performed small RNA seq to investigate underlying molecular mechanism and to reveal functional role of AGO18 in rice. Overall design: We have performed small RNA seq analysis using vegetative and reproductive tissues of both WT and knock-down tansgenic lines. Four weeks old seedlings were considered as vegetative tissues and two different stage of panicle were examined as reprodictive tissues. Libraries were prepared from total RNA isolated from the tissues mentioned.

人工microRNA (miRNA)介导的AGO18基因敲低转基因株系[ago18(1)、ago18(2)]与野生型(WT)对照相比,在营养组织与生殖组织中均表现出发育受损的表型。本研究通过小分子RNA测序(small RNA seq)技术,探究水稻中AGO18的潜在分子机制及其生物学功能。 整体实验设计:本研究对野生型(WT)与敲低转基因株系的营养组织和生殖组织开展小分子RNA测序分析。以4周龄幼苗作为营养组织样本,选取两个不同发育时期的稻穗作为生殖组织样本。测序文库均由上述组织提取的总RNA构建而成。
