Transcriptomic analyses delineated strikingly congruent patterns of gene regulation in hepatocytes stimulated with recombinant activin A and IFNα in vitro. Activin A mRNA, encoded by the INHBA gene, is induced upon activation of RIG-I, MDA5 and TLR7/8 viral nucleic acid sensors in vitro, across multiple cell lines and also in human peripheral blood mononuclear cells Overall design: we first used microarray analysis and RNA sequencing to analyse the effects of activin A upon hepatocytes at the transcriptional level and examine its intersection with the type I IFN axis
转录组学分析(Transcriptomic analysis)清晰揭示:体外条件下经重组激活素A(activin A)与干扰素α(IFNα)刺激的肝细胞,其基因调控模式呈现高度一致的特征。由INHBA基因编码的激活素A mRNA,可在体外多种细胞系及人外周血单个核细胞中,因RIG-I、MDA5与TLR7/8等病毒核酸传感器的激活而上调表达。实验整体设计:本研究首先通过微阵列分析(microarray analysis)与RNA测序(RNA sequencing)技术,在转录组水平分析激活素A对肝细胞的调控作用,并探究其与I型干扰素通路(type I IFN axis)的交叉关联。
