These libraries were used to determine genes that become essential in S. aureus in the absence of the lipid-modifying enzyme MprF. Genes in which transposon reads are significantly depleted in the dMprF library relative to the wildtype library don't tolerate transposon insertions when MprF is deleted and are therefore required for growth in dMprF. Knowledge of these genes can then be leveraged in chemical and genetic screens and selections.
