We used next-generation sequencing to identify differential microRNA (miRNA) signatures in exosomes isolated from ovarian cancer cells and ovarian cancer-associated fibroblasts (CAFs) and adipocytes (CAAs). We found that a significantly higher levels of miR21 isomiRNAs in exosomes isolated from CAFs and CAAs than in those from ovarian cancer cells. Overall design: Samples are exosomes isolated from four ovarian cancer cell lines (A2780, HeyA8, OVCA433 and SKOV3), two normal fibroblasts, three CAFs, two normal adipocytes and two CAAs

我们采用下一代测序技术,对卵巢癌细胞、卵巢癌相关成纤维细胞(cancer-associated fibroblasts, CAFs)及卵巢癌相关脂肪细胞(cancer-associated adipocytes, CAAs)所分离得到的外泌体(exosomes)中的差异微小核糖核酸(microRNA, miRNA)表达谱进行鉴定。研究发现,从CAFs与CAAs分离的外泌体中,miR-21异构体(isomiRNAs)的表达水平显著高于卵巢癌细胞来源的外泌体。总体实验设计:所用样本为外泌体,分别分离自4株卵巢癌细胞系(A2780、HeyA8、OVCA433及SKOV3)、2株正常成纤维细胞、3株CAFs、2株正常脂肪细胞及2株CAAs。
