To determine the impact of infection on NK cells, splenocytes were isolated from spleens of naive or MCMV-infected mice at day 21 p.i. Following isolation, splenocytes were labeled with anti-CD45, anti-CD3, anti-CD19, anti-NK1.1, and anti-DX5 antibodies, and NK cells defined as CD45+, CD3-, CD19-, NK1.1+, and DX5+ cells, were separated from the mixture using fluorescence-activated cell sorting on FACSAriaIIu.
为明确感染对自然杀伤细胞(NK cells)的影响,研究人员于感染后第21天(post infection, p.i.)从未致敏或小鼠巨细胞病毒(MCMV)感染小鼠的脾脏中分离脾淋巴细胞(splenocytes)。分离获得脾淋巴细胞后,采用抗CD45、抗CD3、抗CD19、抗NK1.1及抗DX5抗体对其进行标记;随后借助FACSAriaIIu型流式细胞分选仪,通过荧光激活细胞分选术(fluorescence-activated cell sorting, FACS)从混合细胞群中分离得到表型为CD45+、CD3-、CD19-、NK1.1+、DX5+的自然杀伤细胞。
