Role of alternative polyadenylation in muscle development remains largely undetermined. Our WTTS-seq approach to capture 3''-end of RNAs clearly revealed alternaitve polyadenylation events responsible for reduced muscle mass in AMPK a1, but increased muscle mass in AMPK a2 knockout mice. Overall design: Total RNA samples obtained from muscle tissue of knockout (KO) mice and wild-type (WT) mice were used to construct WTTS-seq libraries. Eight WTTS-seq libraries including two AMPKa1 KO mice, two AMPK a2 KO mice, two AMPKa1 WT mice and two AMPK a2 WT mice were involved in this submission.
可变多聚腺苷酸化(alternative polyadenylation)在肌肉发育中的作用迄今尚未完全阐明。我们所采用的全转录组3'端测序(WTTS-seq)技术可精准捕获RNA的3'末端,该技术清晰揭示:可变多聚腺苷酸化事件是造成腺苷酸活化蛋白激酶α1亚基(AMPKα1)敲除小鼠肌肉量减少、腺苷酸活化蛋白激酶α2亚基(AMPKα2)敲除小鼠肌肉量增加的核心诱因。
整体实验设计:本研究以敲除(KO)小鼠与野生型(WT)小鼠的肌肉组织总RNA为材料构建WTTS-seq文库。本次提交共包含8个WTTS-seq文库,分别为2例AMPKα1敲除小鼠样本、2例AMPKα2敲除小鼠样本、2例AMPKα1野生型小鼠样本及2例AMPKα2野生型小鼠样本。
