In this study, we have developed a novel time to event approach to orodental toxicity prediction, thus providing a more comprehensive estimation of the disease trajectory in order to allow effective risk stratification and surveillance strategies. The developed Weibull AFT model offers a significant advancement in predicting mandibular ORNJ risk following radiotherapy in HNC patients. Being able to predict the time to ORNJ allows for early identification and proactive management of high-risk patients with potential reduction of the severity of ORNJ and improvement of patient outcomes and quality of life.

本研究构建了一种全新的口腔牙齿毒性(orodental toxicity)事件发生时间预测方法,可对疾病进展轨迹开展更为全面的评估,进而为制定有效的风险分层与监测策略提供依据。所开发的威布尔加速失效时间模型(Weibull AFT model),在预测头颈部癌(Head and Neck Cancer, HNC)患者放疗后下颌放射性颌骨骨坏死(osteoradionecrosis of the jaw, ORNJ)风险方面实现了显著突破。通过预测ORNJ的发生时间,能够早期识别高危患者并采取主动干预措施,有望减轻ORNJ的病情严重程度,改善患者的临床预后与生活质量。
