The goal of the study is to compare transcriptome profile of mouse liver with acetaminophen toxicity to potentially therapeutic healthy blood serum exosome or cytokine treatments along with acetaminophen Studies also included sequencing of miRNA from exosomes isolated from pooled serum of several healthy anonimous human donors and livers of mice that received the exosome treatment along with acetominophen. Overall design: Exosomal and tissue small RNAs and tissue total RNA from healthy and injuried states were used to perfom high throughput sequensing by Ion Torrent Proton instrument
本研究旨在对比对乙酰氨基酚(acetaminophen,又名扑热息痛)诱导肝毒性的小鼠肝脏转录组谱,与同时接受对乙酰氨基酚及潜在治疗性健康人血清外泌体(exosome)或细胞因子(cytokine)处理的小鼠肝脏转录组谱。本研究同时包含两类测序分析:一是对多名匿名健康人类供体混合血清中分离得到的外泌体miRNA进行测序;二是对接受外泌体治疗联合对乙酰氨基酚处理的小鼠肝脏样本进行测序。整体实验设计:采集健康状态与损伤状态样本的外泌体、组织小RNA及组织总RNA,通过Ion Torrent Proton测序平台完成高通量测序。
