To explore how p95HER2 regulates gene expression to promote tumor growth in immunocompetent mice, we engineered PB2 mouse breast cancer cells to express p95HER2 under the control of doxycycline. Both empty vector (EV) and p95HER2 cells were implanted to the mammary fat pads of immunocompetent C57BL/6 mice, which were placed under a DOX625 doxycycline diet. Tumor xenografts were harvested 3 weeks after inoculation. We then performed transcriptomic analysis using the RNA-seq data from PB2-EV and PB2-p95HER2 tumors. Comparative gene expression profiling analysis of RNA-seq data from PB2-EV and PB2-p95HER2 tumors.

为探究p95HER2如何调控基因表达以促进免疫健全小鼠的肿瘤生长,本研究通过基因工程手段构建PB2小鼠乳腺癌细胞系,使其在多西环素(doxycycline)的诱导调控下表达p95HER2。将空载载体(empty vector, EV)组细胞与p95HER2组细胞分别接种至免疫健全C57BL/6小鼠的乳腺脂肪垫,随后给小鼠饲喂DOX625规格的多西环素饲料。接种3周后收集肿瘤异种移植物,利用PB2-EV与PB2-p95HER2肿瘤的RNA-seq数据开展转录组分析,并对两组肿瘤的RNA-seq数据进行比较基因表达谱分析。
