Inhibitors of glucose (IO+DHEA group) or fatty acids (ETOMOXIR group) metabolism were applied during bovine oocyte in vitro maturation (IVM). Control group was conducted in standard maturation conditions. In vitro fertilization and embryo culture were applied. Obtained blastocysts were analysed with regard to lipidome, metabolome (mass spectrometry), transcriptome (RNA Seq) and lipid droplets staining (BODIPY). Inhibitors of glucose (IO+DHEA group) or fatty acids (ETOMOXIR group) metabolism were applied during bovine oocyte in vitro maturation (IVM).
在牛卵母细胞体外成熟(in vitro maturation, IVM)过程中,分别施加葡萄糖代谢抑制剂(IO+DHEA组)与脂肪酸代谢抑制剂(ETOMOXIR组)。对照组采用标准成熟培养条件开展实验。随后进行体外受精与胚胎培养操作,对获得的囊胚进行分析,分析维度包括脂质组学(lipidome)、代谢组学(mass spectrometry,质谱法)、转录组学(transcriptome,RNA Seq)以及脂滴染色(BODIPY)。在牛卵母细胞体外成熟(IVM)过程中,分别施加葡萄糖代谢抑制剂(IO+DHEA组)与脂肪酸代谢抑制剂(ETOMOXIR组)。
