In previous studies we identified the small RNA-binding protein CcaF1 that is involved in sRNA maturation and RNA turnover in Rhodobacter sphaeroides under microaerobic growth conditions (Grützner et al., 2021, Nucleic Acids Res. 49(6), doi: 10.1093/nar/gkab146). In this study we analysed the the small RNA-binding protein CcaF1 under phototrophic growth condtions to identify new RNA binding partners. Overall design: The CcaF1 protein was overexpressed with an 3xFLAG (pRKccaF1FLAG) in R. sphaeroides under phototrophic growth condtions
前期研究中,我们在微好氧生长条件下的球形红杆菌(Rhodobacter sphaeroides)中鉴定出一种参与sRNA(small RNA)成熟与RNA降解的小RNA结合蛋白CcaF1(Grüutzner等人,2021,《核酸研究》(Nucleic Acids Research),49(6),doi: 10.1093/nar/gkab146)。本研究针对光养生长条件下的小RNA结合蛋白CcaF1开展分析,以鉴定其新型RNA结合伴侣。实验整体设计:将带有3×FLAG标签的CcaF1蛋白(质粒pRKccaF1FLAG)在光养生长条件下的球形红杆菌(R. sphaeroides)中进行过表达。
