This work investigated the effect of High-fat diet (HFD) on mouse brain cortices, in APOE3 and APOE4 background. We hypothesized that HFD-induced obesity alters brain transcriptomics in APOE-genotype and sex dependent manners. Six-month-old APOE3-TR and APOE4-TR mice were treated with either HFD or control chow. After 4 months, total RNA was extracted from the cerebral cortices and analyzed by poly-A enriched RNA sequencing on the Illumina platform. Here, we investigated interactions between HFD, APOE, and sex, using a knock-in mouse model of the human APOE3 and APOE4 alleles.
本研究探究了高脂饮食(high-fat diet, HFD)对载脂蛋白E3(APOE3)与载脂蛋白E4(APOE4)遗传背景下小鼠大脑皮层的影响。我们提出假设:高脂饮食诱导的肥胖会以APOE基因型和性别依赖的方式改变大脑转录组特征。选取6月龄的APOE3靶向替换(APOE3-TR)与APOE4靶向替换(APOE4-TR)小鼠,分别给予高脂饮食或对照饲料干预。干预4个月后,提取小鼠大脑皮层的总RNA,并通过聚腺苷酸富集RNA测序(poly-A enriched RNA sequencing)在Illumina测序平台完成分析。本研究借助人源APOE3与APOE4等位基因的敲入小鼠模型,探究了高脂饮食、APOE基因型与性别三者间的交互作用。
