We performed tagmentation-based whole-genome bisulfite sequencing on young and aged LT-HSCs. Overall design: Tagmentation-based whole-genome bisulfite sequencing (T-WGBS) of aged LT-HSCs (5 Replicates) and young LT-HSCs (4 Replicates). For each replicate, two separate T-WGBS libraries were created and sequenced on one lane per library.

我们对青年与老年长期造血干细胞（long-term hematopoietic stem cells, LT-HSCs）开展了基于转座酶介导的全基因组亚硫酸氢盐测序（tagmentation-based whole-genome bisulfite sequencing）。实验整体设计：采用基于转座酶介导的全基因组亚硫酸氢盐测序（T-WGBS）技术，检测样本为老年LT-HSCs（5个生物学重复）与青年LT-HSCs（4个生物学重复）。针对每个生物学重复，均构建两份独立的T-WGBS文库，每份文库单独在一个测序泳道中完成测序。
