We used single-cell RNA sequencing to analyze immune cells in COVID-19 patients. We drew blood from convalescent patients and examined PBMCs, monocytes and regulatory T cells. Monocytes and Tregs were sorted using fluorescence activated sorting (FACS). All samples were sequenced using a 10X genomics library preparation kit and an Illumina HiSeq sequencer.
本研究借助单细胞RNA测序(single-cell RNA sequencing)技术,针对新型冠状病毒肺炎(COVID-19)患者体内的免疫细胞开展分析。我们从康复期患者体内采集血液样本,对外周血单个核细胞(peripheral blood mononuclear cells, PBMCs)、单核细胞及调节性T细胞进行检测。其中单核细胞与调节性T细胞(Tregs)通过荧光激活细胞分选术(fluorescence activated sorting, FACS)完成分选。所有样本均采用10X Genomics文库制备试剂盒与Illumina HiSeq测序仪完成测序。
