Ovarian cancer (OC) is the leading cause of death from gynecologic malignancies in the US. Ovarian cancer stem cells (OCSCs) have been shown to drive chemoresistance and tumor progression but the mechanism remains incompletely understood. Aldehyde Dehydrogenase 1A1 (ALDH1A1) is a robust marker for cancer stem cells in ovarian and other cancers. We demonstrate that ALDH1A1 inhibition suppresses stemness, chemoresistance and senescence in ovarian cancer. Overall design: In triplicate, OVCAR3 cells were treated with compound 974 (5µM for 48h) or DMSO. RNA was harvested from the cells using the RNAEasy kit (Qiagen,Catalog No; 74106). RNA-seq libraries were prepared using TruSeq Stranded mRNA Library Prep Kit (Illumina #RS-122-2101) according to manufacter's protocol.
卵巢癌(Ovarian cancer, OC)是美国妇科恶性肿瘤相关死亡的首要病因。卵巢癌干细胞(Ovarian cancer stem cells, OCSCs)已被证实可驱动化疗耐药与肿瘤进展,但其具体作用机制尚未完全阐明。乙醛脱氢酶1A1(Aldehyde Dehydrogenase 1A1, ALDH1A1)是卵巢癌及其他癌症中癌症干细胞的可靠标志物。本研究证实,抑制ALDH1A1可抑制卵巢癌的干细胞干性、化疗耐药及细胞衰老。实验设计:以三组生物学重复的方式,将OVCAR3细胞用化合物974(5µM处理48小时)或二甲基亚砜(DMSO)处理。采用RNAEasy试剂盒(Qiagen,货号:74106)从细胞中提取总RNA。按照制造商操作规程,使用TruSeq链特异性mRNA文库制备试剂盒(Illumina #RS-122-2101)构建RNA测序文库。
