High-throughput nanopore DNA sequencing of large insert fosmid clones directly from bacterial colonies

Establishment of a method to sequence 96 fosmid in a single sequencing run using Oxford Nanopore Technologies. The sample preparation involves fosmid amplification from Escherichia coli clones using phi29 polymerase and then direct nanopore sequencing using the Oxford Nanopore Technologies system.

本研究建立了一种可利用牛津纳米孔科技（Oxford Nanopore Technologies）平台单次测序运行即可完成96个黏粒（fosmid）测序的方法。该方法的样本制备流程包括：采用phi29聚合酶（phi29 polymerase）从大肠杆菌（Escherichia coli）克隆菌株中扩增黏粒，随后借助牛津纳米孔科技测序系统开展直接纳米孔测序。
